所谓自由基(free radical)简单来说就是在外围拥有不成对电子的原子或是分子.我们知道原子是由原子核及外围的电子所构成的,在稳定情况下,这些外围的电子应该是成对的.当因某些原因使得原子或是分子失去或是多获得一个电子时,此原子或是分子的外围就拥有了不成对的电子,而形成了所谓的自由基.自由基是一种极稳定的状态,它随时想从周围的其他原子或是分子处掠夺或是推送一个电子,目的是使自己尽快回复到稳定状态.我们体内的自由基在掠夺或是推送电子时,可能会对其周围的细胞,组织,器官或是外来的细菌,病毒,及各种病原体造成伤害. 生物内最常见的自由基是氧自由基(free oxygen radical),这些氧自由基主要是由细胞内的粒线体(mitochondria)所产生的.粒线体是相当於生物体的能源工厂,一切生命现象所需的能源都来自粒线体.粒线体藉由电子传递链(electrode transport chain)的机转来产生能量,而氧原子是此电子传递链的最后一棒.所以免不了在产能过程中会产生一些氧自由基出来.换句话说,只要有生命现象的存在,就免不了氧自由基的产生,所以我们是不可能完全消除自由基的,除非生命现象已停止. 根据自由基理论 (Free radical theory), 发生在生物体的疾病及老化有一大部和氧自由基的存在有关.所以,生老病死是宿命的过程.事实上,我们的身体已经有了一套抗氧化系统,包括一些酵素(如:catalase,glutathione,peroxidase)及一些抗氧剂(如:Vit C,Vit E及beta-胡萝卜素),他们可帮忙清除这些氧自由基.在人体和环境中持续形成的自由基来自人体正常新陈代谢过程，大量体育运动、吸烟、食用脂肪和腌熏烤肉、发生炎症、某些抗癌药物、安眠药、射线、农药、有机物腐烂、塑料用品制造过程、油漆干燥、石棉、空气污染、化学致癌物、大气中的臭氧等也都能产生自由基。已知自由基可损伤蛋白质，可使蛋白质的转换增加；损害DNA可导致细胞突变；作用于－SH可使某些酶的活性降低或丧失；攻击未饱和脂肪酸可引起脂质过氧化，其氧化产物可引起－SH氧化、酶失活、膜功能受损、干扰膜的运送功能等。另外，由燃料废气、香烟和一些粉尘造成的大气污染，使大气上空的自由基占分子污染物总量的1％～10％，因此环境污染中的自由基反应也是不可忽视的
